Molecular markers are important tools in the characterization of plant genetic diversity and can provide support for conservation strategies endangered populations. The different molecular techniques involve evaluation many individuals; therefore, it is crucial to have fast, efficient, inexpensive methods DNA extraction. Given importance Aroeira (Myracrodruon urundeuva Fr. All.) pertinent optimize a protocol that allows obtainment intact pure DNA, aiming assist this species threatened with extinction. Thus, study aimed compare five extraction methods: Dellaporta et al. (1983), Doyle (1987) modified, Ferreira Grattapaglia (1995), Romano Brasileiro (2015), Khanuja (1999) most efficient M. urundeuva. modified proposed by (1987), using 100 mg leaf tissue 6 µl β-mercaptoethanol was presented sharpest bands after electrophoresis reactions amplification employing Polymerase Chain Reaction (PCR).
